Epigenetics

Reduced Representation Bisulfite Sequencing (RRBS)

Enrich CpG-rich regions to investigate DNA methylation using a focused sequencing strategy.

Principle

RRBS enriches CpG-rich regions through restriction digestion and fragment selection, followed by bisulfite treatment, amplification and sequencing. It focuses methylation comparisons on covered promoters, CpG islands and other selected regions.

Applications

  • Study methylation during differentiation and development
  • Compare CpG-rich regions associated with disease or treatment
  • Assess drug-related methylation changes within covered regions

Method features

  • Single-base resolution within the achieved coverage
  • Focuses data on CpG-rich regions
  • Project design considers sites jointly covered across samples

Workflow

  1. 01

    Extract and digest DNA

  2. 02

Select fragments and perform bisulfite conversion

  3. 03

    Amplify, assess and sequence libraries

  4. 04

Quantify and compare methylation within covered regions

This is a workflow overview. Experimental and analytical details depend on sample conditions and research objectives.
